Biotools Enzymes for PCR

  • Below is a brief description of Biotools enzymes for PCR and thermocycle-related applications.

    To improve storage and shipment, and make handling easier, Biotools provide its enzymes in its patented "gelification" format, as well as in a standard enzyme mix. Gelfication allows Biotools to provide enzymes as ready-to-use one-tube pre-made reaction mixes, so the researcher simply needs to add sample and water. In addition to convenience, this format enables hot-start PCR since the component will not react in the gel, unless the temperature is above 90 C. This format makes Biotools enzymes more convenient for the researchers and facilitates automation, as well as minimizing contamination, procedural errors, and mis-priming during PCR.

    Biotools Thermostable DNA Polymerase --- recombinant DNA polymerase from Thermus thermophilus HB27.

    • 5' to 3' polymerase activity, and weak 5' to 3' exonuclease activity.
    • No detectable 3' to 5' exonuclease activity (it is not a proof-reading enzyme).
    • No reverse transcriptase activity.
    • Average error rate is 1 in 10 kbp (Taq error rate 1 in 6 kbp)
    • The enzyme half life at 94 ºC is 40 minutes. This means that the polymerase still retains most of its activity even after 100 amplification cycles.
    • Larger active site than Taq polymerase so large nucleotides such as biotin, fluorescein and digoxigenin-labeled dNTPs (including dUTP), as well as radioactively labeled dNTPs, can be used efficiently as substrates.
    • Terminal transferase activity, so amplification products can be directly used for T/A cloning.

    Retrotools RT-PCR Kit --- a derivative of BIOTOOLS DNA Polymerase that has been protein-engineered to enable reverse transcriptase activity.

    • Reverse transcriptase activity, as well as 5' to 3' polymerase activity, and weak 5' to 3' exonuclease activity.
    • No detectable 3' to 5' exonuclease activity (it is not a proof-reading enzyme).
    • Use one enzyme for the whole reverse-transcriptase-PCR process for products up to 1 Kbp.
    • Reverse transcription is performed at high temperatures (65 ºC or even 70 ºC) which minimizes the formation of RNA secondary structure that can inhibit efficient and complete transcription.

    Biotools Pfu --- thermostable recombinant DNA polymerase from Pyrococcus furiosus

    • 5’ to 3’ polymerase activity,
    • 3’ to 5’ exonuclease proofreading activity.
    • Error rate 10-fold lower than non-proofreading enzyme (1 x 10-6).
    • Amplifies up to 13 Kbp.
    • Amplification products can be directly used in blunt-ended vectors.
    • Radioactively labelled dNTPs, as well as biotin, fluorescein, and digoxigenin-labelled dNTPs can be used as substrates.

    Certamp PCR Enzyme Kits --- a unique blend of enzymes and cofactors that maximize processivity, fidelity and yield, while minimizing errors

    • Each kit contains different enhancing buffers and additives to provide the best results for select applications
    • Optimized kits for applications such as amplification of templates with high percentage of secondary structure, amplification of long templates (from 5 to 40 Kbp), and amplification where template is limiting.

    Biotools Pyrophosphatase --- enhances amplification reactions by removing pyrophosphate, an inhibitory by product remaining of polymerase activity.

    • Simply add it to PCR or sequencing reactions when low band intensity is observed.
    • Improve amplification where the amount of template DNA is limiting, reaction times are long, or any other reason to maximize yield.
