B-Bridge's Human, Mouse, and Rat Resistin ELISA Kits make use of a straightforward biotin/horseradish peroxidase-labeled streptavidin reaction to provide sensitive and quantitative measurements of homodimeric resistin in serum, plasma, or tissue culture medium.

| Assay Description |
|
| Samples and corresponding controls are incubated in microtitration wells coated with a goat anti-human or anti-mouse resistin antibody. After a thorough wash, polyclonal anti-human or anti-mouse resistin antibody, which is labeled with biotin, is added to the wells and incubated with captured resistin. Following another washing step, the streptavidin-horseradish peroxidase conjugate is added. After a one hour incubation and the last washing step, the remaining conjugate is allowed to react with the substrate H202 tetramehtylbenzidine (TMB). The reaction is stopped by addition of an acidic solution and the resulting yellow product is measured at 450 nm. The absorbance is proportional to the concentration of resistin. A standard curve is constructed by plotting absorbance values versus resistin concentrations of calibrators and concentrations of unknown samples are determined using this standard curve. |
